§3.1Obesity and overweight in adults

Anchor outcome. Percentage change in body weight from baseline.

Effect as recorded. −10.8 % body weight at 26 weeks with cagrilintide 4.5 mg monotherapy versus −3.0 % with placebo, and −8.6 % with liraglutide 3.0 mg as active reference; estimated difference versus placebo −7.8 percentage points (95 % CI −10.2 to −5.4).[1,2]

Certainty. Moderate certainty Phase 2 dose-finding, 706 participants. Monotherapy effect is smaller than that of the incretin class; the therapeutic interest lies chiefly in combination.

§3.2Type 2 diabetes mellitus

Anchor outcome. Change in HbA1c (%, mmol/mol).

Effect as recorded. Combination with semaglutide 2.4 mg gave −15.6 % weight and −2.2 % HbA1c at 32 weeks; phase 2; combination arm.[2,3]

Certainty. Low certainty Attribution of effect between components is not resolvable from the reported design.
